Resumo

Dengue viruses are the most important arthropod-borne viruses in terms of morbidity and mortality in the world. Dengue viruses are single-stranded, positive-sense RNA viruses that belong to the genus Flavivirus of the family Flaviviridae. They consist of four serotypes (from DENV-1 to DENV-4) that are transmitted to humans and other higher primates by mosquitoes of the genus Aedes. The only available way to control dengue is vector control since there is neither a vaccine for human use nor an antiviral drug to inhibit virus replication. The present study examined the effect of leaf extracts of Annona muricata on the dengue-2 virus replication in Vero (mammalian) cells in order to define whether or not extract could be used to reduce viral yield. Vero cells were cultivated in a 24-well plate and the confluent monolayers were infected with dengue-2 virus at a multiplicity of infection of 0.1. To analyse the inhibition of viral replication by this extract, monolayers of Vero cells were treated, 1 hour after viral infection and at different intervals of the time (24-24 hours) with extract at concentration of 1mg/mL. Supernatants of infected cells were collected at defined periods of 24, 48, 72, 96, 120, 144 and 168 hours after viral infection, the total RNA was extracted from the supernatant, and viral replication was assessed by Real-time RT-PCR using primers specific to the 3' non-coding region of the dengue-2 virus genome and SYBR GREEN® One-Step RT-PCR Master Mix Reagents Kits (Applied). Our results showed that, compared to untreated cells, there is a statistically significant decrease in viral replication in Vero cells when treated with extract. The extract induced an effective inhibition in dengue-2 virus replication when administered 24 hours after infection in vitro (Vero cells). The study presented preliminary evidences that leaf extracts of Annona muricata interferes with dengue-2 virus replication in Vero cells culture.
