423-1INFLUENCE OF TEMPERATURE ON PRODUCTION OF CELLULASES AND XYLANASE BY Trichoderma STRAINS

Resumo

The solid-state fermentation (SSF) has great potential as a process for the production of enzymes. The agro-industrial residues used for SSF as a low cost substrate is of producing promising as inducers of enzymes such as cellulases and xylanase. The aim of present study was to evaluate the influence of temperature conditions on production of CMCase, &beta-glucosidase and Xylanase by specimens of Trichoderma virens and Trichoderma harzianum. For evaluated the production of enzymes, the specimens of Trichoderma were studied and cultivated using state solid fermentation (SSF) condition in a mixture of sugarcane bagasse and wheat bran (1:1). Conidia or mycelial biomass of cultures with 4 days growth were suspended in nutrient solution Urea (1%, pH5) and the incubation time was during 144h. Four different sources of temperature (28°C, 30°C, 37°C, 39°C) were tested. The production of CMCase was average greatest of 11 U/g (Trichoderma harzianum) and 8 U/g (Trichoderma virens). For xylanase, the results observed for activity enzymatic were 25 U/g (Trichoderma harzianum) and 22 U/g (Trichoderma virens). However, for both strains tested, the production of &beta-glucosidase was less efficient in all incubation temperatures observed, when compared to others enzymes production. Similarly, other studies showed that at lower temperature activity was better when compared to higher temperatures. The enzymes are produced during the exponential growth phase of organism. Incubation at higher temperatures decreases the growth of micro organism and the enzyme production. The specimens were studied CMCase and xylanase producing good. However, for the production of &beta-glucosidase is necessary to conduct further experiments in order to obtain satisfactory results, such tests time incubation, source nitrogen and pH.
